Abstract | 「要旨」N-isopropyl-p-iodoamphetamineの肝集積機序を解明する目的で, Wister系雄性ラット5匹を用いて, 123I-IMP門脈投与による肝動態検査と同系ラット2匹を用いて125I-IMPによる肝ミクロオートラジオグラフィを施行した. 123I-IMP門脈投与により, 123I-IMPは肝内に高率に摂取され, 洗い出しも比較的速やかであった(静注後10分の肺/肝カウント比0.2, 0.15). 塩酸ケタミン5mg, 8mg負荷により123I-IMP肝集積は低下し, 静注直後より肺描出がみられた(静注後10分の肺/肝カウント比0.20). 肝ミクロオートラジオグラフィの検討では, 黒化度は中心静脈および洞様血管内に認められ, 肝実質細胞内には認められなかった. 以上の結果から, 肝内中心静脈の内皮細胞内の非特異的アミン受容体の存在が示唆されたが, 未だ実験動物数が少なく, 今後の研究がまたれる. |

Abstract | In order to clarify the mechanism of N-isopropyl-p-iodoamphetamine (IMP) liver accumulation, liver dynamic study by the portal injection of 123I-IMP and liver microautoradiography by 125I-IMP were performed using 5, 2 male rats, respectively. The initial uptake of 123I-IMP in the liver was very high and thereafter 123I-IMP showed relatively rapid wash-out (count ratio of lung to liver at 10 min after the injection was 0.12, 0.15). On the other hand, the addition of 5 mg, 8 mg ketamine hydrochloride decreased the initial 123I-IMP liver uptake and its lung accumulation was noted immediately after the injection (count ratio of lung to liver at 10 min was 0.20). Microautoradiography of the liver using 125I-IMP showed grain density in the central vein and sinusoids, but not in the liver parenchymal cell. These results suggest that non-specific amine receptor (binding site) may exist in the endothelial cell in the central vein, although the number of experimental rats in this series was small for conclusion. |
