Abstract | 「要旨」動脈血栓症をイムノシンチグラフィの手法により描出する目的で, ウサギおよびヒトの血小板に対するモノクローナル抗体を作成しその性状の検討を行った. ウサギ血小板に対して12種, ヒト血小板に対しては2種の抗体が確立された. これらはすべて血小板特異的で, 血小板の凝集能に影響を及ぼすものが多かった. これらのなかでウサギ血小板を抗原として作られた138-10は1.2×10 9M-1の高い親和定数でウサギ血小板と結合し, ヒトの血小板に対して作られた抗体1-19の親和定数は2.1×10 9M-1であったが, ウサギの血小板ともほぼ同様の親和定数で結合した. また, これら二つの抗体はともに血小板の凝集能にほとんど影響を及ぼさなかった. 138-10と1-19はその高い親和定数とウサギ血小板凝集能に影響を与えない特性から, ウサギを用いた血栓シンチグラフィの基礎検討に応用可能と思われた. |

Abstract | Intending to use for the immunoscintigraphy (IS) of arterial thrombosis, we have generated monoclonal antibodies reactive with rabbit and human platelets. Twelve monoclonal antibodies were established by the immunization of rabbit platelets and two by human platelets. All antibodies were specific for platelets, but some recognized both human and rabbit platelets. Antibody, designated 138-10, bound only to rabbit platelets with a high association constant of 1.2×10 9M-1 and showed little effect on the aggregation of platelets. Another antibody 1-19 was produced by the immunization of human platelets but cross-reacted with rabbit platelets. As far as we know, this is the first report describing about monoclonal antibodies reactive with rabbit platelets, and these radio-labeled monoclonal antibodies will be of great use for the study of thrombus scintigraphy. |
