Abstract | 「要旨」著者が作成したγ-seminoprotein(γ-Sm)に対するモノクローナル抗体(IgG1, K)およびそのF(ab')2フラグメントを125I標識して, 担ヒト前立腺腺腫ヌードマウスにおける体内動態を観察した. 125I-intact IgGおよび125I-F(ab')2投与後96時間の全身オートラジオグラムでは, いずれによっても腫瘍部に良好な集積を認めた. 組織内放射能濃度測定でも腫瘍部は比較的高値を示した. 抗γ-Sm MoAbによる前立腺腫瘍イムノシンチグラフィへの足掛かりとしたい. 「I. はじめに」1966年, 原らは精漿の免疫電気泳動において, γ-globulin域に特異成分を見いだし, γ-seminoprotein(以下γ-Smと略)と命名した. γ-Smは約10%の糖を含む分子量約29,000の糖蛋白で, ヒト前立腺に特異的に見いだされ, その臓器特異性および種属特異性が知られている. 免疫組織化学的には, 正常前立腺, 前立腺過形成, そして前立腺癌組織いずれにおいても, その腺上皮細胞および腺腔内にγ-Smの局在が認められる. |

Abstract | [Summary]The biodistribution of 125I labeled monoclonal antibody against gamma seminoprotein (γ-Sm) in the nude mice bearing human benign prostatic hyperplasia xenograft was evaluated by whole body autoradiography and by counting of radioactivity in organs. The monoclonal antibody (murine Ig G1, K) to γ-Sm which was established in this institute and its F(ab')2 fragment were radioiodinated using Iodogen method. The autoradiograms demonstrated specific uptake of 125I-intact Ig G as well as 125I-F(ab')2 within the prostatic adenoma xenografts 4 days after intravenous administrations. Radioactivity in the xenograft was relatively higher than those in the liver, kidney or lung. These results suggest that radio-labeled monoclonal antibody against γ-Sm might be applicable for radioimmunodetection of prostatic tumors which produce γ-Sm. |
