Abstract「要旨」 アドリアマイシン(ADR)心筋障害ラットを用いて, 心筋疾患のミトコンドリア機能評価におけるヨード脂肪酸(125I-BMIPP)の有用性を検討した. ADR 4mg/kg 6日間の連続投与で電顕的にはミトコンドリアの変性, 膨化が認められ, また生化学的にもミトコンドリア呼吸機能の低下が確認された. しかし, 筋原線維, 核などには明らかな障害は見られず, 心筋細胞径, 線維化率にもADR群と対照群との間に差はなかった. また201TlClの心筋内摂取率にも両群間で差はなく, 今回のADR群では明らかな心筋灌流, 心筋線維化や, Na-K ATPase活性などの膜機能障害は見られず, かなり特異的なミトコンドリア障害を作製し得たものと考えられた. これに対して, 125I-BMIPPの心筋内摂取率はADR群で26%有意に低下し, これは主にミトコンドリア(31%), 上清(26%)分画に見られた. したがって本モデルでは, ミトコンドリア機能障害を反映して125I-BMIPPの心筋内摂取が低下したものと考えられ, 本核種は心筋疾患患者の脂肪酸代謝, ミトコンドリア障害を検出する核種として有望なものと推測された.

Abstract[Summary] The ability of 125I-BMIPP in assessing myocardial mitochondrial function was investigated in rats treated with adriamycin (ADR). Intraperitoneal administration of ADR (4 mg/kg) for 6 days significantly decreased a ratio of heart/body weight as compared with the control rats. The light and electron microscopic analysis showed that ADR produced histological damages mainly in the mitochondria with minimal changes in the myofibril and nucleus. In the study of mitochondrial respiratory function, oxygen consumption with adding ADP, respiratory control ratio and ADP/O ratio were significantly lower in ADR rats than control rats. While myocardial uptake of 201TlCl was not different between the two groups, the uptake of 125I-BMIPP corrected by blood counts was significantly reduced in ADR rats (3.1+-0.4 vs. 2.3+-0.8, P<0.05) and the reduction was mainly observed in the mitochondrial fraction. These results suggest that impairment of mitochondrial function by ADR produced a reduction in mitochondrial and consequently myocardial uptakes of 125I-BMIPP. The observation suggests that 125I-BMIPP myocardial scintigraphy could be a useful method for determining mitochondrial function in patients with myocardial diseases.
