Abstract | 「I. 緒言」 各種臓器スキャンニング用放射性医薬品は, 現在のところ99mTc標識化合物が主体である. 当然, そのほとんどが水溶性であり, かつ短半減期, γ線放出物質である. これら放射性医薬品の正確で微細な臓器内分布を解明することは各種臓器スキャンの読影の裏付けともなり, きわめて重要かつ有用なことと考えられる. しかしこの解析手段としてミクロオートラジオグラフィー (以下オートラジオグラフィーをARGと略す) を用いる場合, 上記のように使用医薬品の物理的あるいは化学的性質を考慮すると, きわめて技術的な困難さが予想される. これらの問題を充分考慮した上で, 著者らは今回まず, 腎スキャンニング剤, 99mTc-DTPAをとりあげ, 本剤の細胞レベルでの腎内動態を明らかにする目的で, 小動物を用いてミクロARGを試み, 各種技術的検討を重ね, 初期の目的を達成できたので, 方法論を中心に述べたいと思う. |

Abstract | [Summary] Renal localization of 99mTc-DTPA (diethylenetriamine penta acetic acid) 2 minutes after injection into rats, was investigated using microautoradoigraphy. Besides, optimal technical conditions for the autoradiography with 99mTc were also studied. Because this radionuclide is water soluble substance with short physical half-life, peculiar methods were requireed. For preventing diffusion of soluble radionuclides until exposure to the autoradiographic emulsion, frozen sectioning and frozen exposure methods were accepted in the dark room. Besides, minimizing the decrease of radioactivity, some modifications against the usual procedure had to be done. In spite of these disadvantages, we could get favourable result by these methods about the information on the renal distribution of 99mTc-DTPA. Microautoradiographs demonstrated the specific accumulation to the glomeruli at 2 minutes after injection. There were little accumulation in the tubules and collecting ducts. |
