Abstract「要旨」化学発癌剤, 3'-Methyl-4-dimethylaminoazobenzene(3'-Me-DAB)連続投与ラットについて, 1週から20週までの67Ga-citrate(67Ga)の肝摂取率変化を検索した. 67Gaの肝摂取率は, 3'-Me-DAB投与開始後3週目で一過性の上昇を示し, その後一旦低下するが6週目より再び上昇し, 20週目における67Gaのg肝あたりの摂取率は対照群の約2.3倍の値を示した. この67Gaの肝摂取率パターンは, 肝γ-glutamyltranspeptidaseやglucose-6-phosphate dehydrogenase酵素活性の変動と一部関連が認められた. さらに, 67Gaの細胞内分布を経時的に追跡比較した結果, 800×9分画に著しい変化が認められた. これらの結果から, 3'-Me-DABにより誘導される病変に関与するものの一つに800×g分画に含まれる成分が考えられ, またこの成分が67Gaとの結合性に関与していることが示唆された.

AbstractThe changes of 67Ga-citrate(67Ga)in the rat liver during feeding with chemical carcinogen 3'-methyl-4-dimethylaminoazobenzene(3'-Me-DAB)were studied for 20 weeks. The results were shown as follows: Elevated 67Ga accumulation in the rat liver was first observed at 3rd week after the start of 3'-Me-DAB feeding. There was decrease from the 3-week level at about the 6th week, followed by a sustained increase. The accumulation of 67Ga in the gram liver at 20th week was about 2.3 times higher than that of the control. There was close correlation between the 67Ga accumulation pattern and the patterns of the hepatic γ-glutamyltranspeptidase and glucose-6-phosphate dehydrogenase activities at late stages during hepatocarcinogenesis. In subcellular distribution of 67Ga, remarkable changes were seen in 800×g fraction. From these studies, it is suggested that 67Ga may bind with components in 800×g fraction, concerned with one pathological changes induced by 3'-Me-DAB.
