Abstract | 「要旨」111In-オキシンの合成, および111In-オキシンを使った111In-標識白血球の調製について, 基礎的検討を行ない, 炎症巣のin vivo検出を試みた. オキシンは, pH 5〜6の条件下, 50〜100 μgで, 20μCi〜1 mCiの111Inに対して, 70〜90%の111In-オキシン収率を与えることがわかった. この111In-オキシンによる白血球標識は, 〜10 6の白血球に対しては血球数に比例して増加する. また, この標識白血球の数は, インキュベーション時間に依存して, 非直線的に増加するが, それらの標識111Inのなかで, 血漿との相互作用後も, 安定に血球中に残るものは, 時間とともに比較的ゆっくりと, 直線的に増加していくことがわかった. 動物を使った炎症のin vivo検出では, 静注された111In白血球は, 炎症巣に集積して, その像を描出することができた. しかし一方, 失敗例から, 操作の上での白血球の失活は, 最も大きな問題であることがわかった. |

Abstract | [Summary] : Basic studies on chelation of 111In by oxine and on labeling of leucocytes with the chelate, 111In-oxine, were performed. Fifty to hundred μgs of oxine made chelate complexes with 20μCi-1mCi of 111In, giving 70-90% chelation efficiency. Using these 111In complexes, leucocytes were successfully labeled : the leucocytes tagged with 111In increased in proportion to the number of cells added. The 111In-oxine molecules which remained stable in the cells when incubated with plasma at 37°C, showed linear and rather slow increase depending on incubation time, while whole amount of labeling 111In increased nonlinearly and faster. These stable 111In-oxine molecules are considered to be fixed in the cell membrane structure. The 111In-leucocytes injected intravenously accumulated into the abscess and gave a clear scintigraphic image in animal studies. However, some unsuccessful studies revealed that the labeling procedure produces impaired leucocytes which have lost the ability to function as normal ones. |
