Abstract「要旨」99mTcによる血小板標識に関する検討を, CIS製99mTc赤血球標識用キット (TCK-11) を用いて検討した. 標識用キットで処理した血小板を99mTcを加えてインキュベートすると90分で最大11.8±0.2% (n=3) の標識率が得られ, 連続8回までの洗浄では99mTcの血小板よりの溶出は認められなかった. これらの結果をもとにして被検者の血液200mlより分離した血小板を99mTc 2mCi, 51Cr 500μCiにて標識し, 各種疾患11例で血小板の臓器分布をシンチレーションカメラにより観察し, 併せて血小板寿測定も行った. その結果, うっ血性脾腫の例では脾への99mTc標識血小板の強い取り込みを認め, 特発性血小板減少性紫斑病の急性期例では血小板寿命の短縮, 交替率の亢進, 脾への選択的取り込みを認めた. 99mTc, 51Cr二重標識法は, 従来の51Cr法に比し, 可視的に血小板の抑留破壊臓器をとらえることができ, 有力な情報を提供するものと考えられた.

Abstract[Summary] In vitro studies of platelet labelling with 99mTc by using CIS kit for 99mTc red blood cell label were done inorder to standardize the technique of platelet labelling. These results were applied to in vivo clinical studies and organ distribution of 99mTc-labeled platelets was observed by scintillation camera in various hematological disorders. This method was considered to be especially useful in some disorders such as idiopathic thrombocytopenic purpura in which platelets were destructed by antiplatelet antibodies and congestive splenomegaly which has increased splenic pooling. Additionally, these technique are excellent because platelet survival could also be obtained by labelling platelets with 51Cr together with 99mTc.
