Abstract「要旨」血清中の3,3'diiodothyronineの測定法としてのradioimmunoassay法を確立したので報告する. 抗3,3'diiodothyronine血清はL-3,3'diiodothyronineをカルボデイミドを用いて牛血清アルブミンに結合し, この結合物を抗原としてcomplete Freund Adjvantと共に家兎に感作して作成した. この抗血清は甲状腺ホルモン及びその誘導体とは免疫交叉性を示さず, 3,3'diiodothyronineに特異的抗血清であることが認められた. boundとfree formの分離には二抗体法を用いた. 本法における最低測定感度は0.5ng/dlであった. 回収率, intraassay reproducibility, interassay variationも良好であった. この方法で測定した人血清中の3,3'diiodothyronine値は正常者で平均6.4ng/dl, 甲状腺機能亢進症で高値, 甲状腺機能低下症で低値であった. 以上本法は血清中の3,3'diiodothyronine測定法として十分用い得る方法であることが認められた.

Abstract[Summary] A sensitive, specific and reproducible radioimmunoassay for measurement of 3,3'-L-diiodothyronine (T2) in unextracted human serum is described. The binding antiserum was prepared by immunization of rabits with T2-bovine serum albumin conjugate. This antiserum did not cross-reacted significantly with various thyroid hormone derivatives. Assay sensitivity was considered to be 0.5 ng/dl by stastical criteria. The mean recovery of different concentrations was 101%. Intraassay reproducibility averaged and interassay variation were 4.0%, 6.4%, respectively. 8-anilino-1-naphthalene sulfonic acid (200 ug/tube) was used to inhibit T2 to binding to serum protein. The mean (Mean+SD) serum T2 concentration was 6.4+3.3 ng/dl in normal subjects, 31+18 ng/dl in hyperthyroidism, 3.2+2.5 ng/dl in hypothyroidism. T2 levels in serum were decreased to normal range in patients with hyperthyroidism during antithyroid drug treatment. T2 levels in serum were increased to normal range in patients with hypothyroidism during thyroxine treatment. These data suggest that this assay system renders attractive for clinical determination of T2 in human serum.
