Abstract | 「要旨」: 99mTc標識肝胆道系スキャン物質の胆道系排泄機能について検討するため, sulfobromophthalein (BSP) の最大排泄状態 (Tm) 下にあるラットに99mTcHIDAを静注し, 血漿クリアランス, 胆汁内回収率を測定し, 対照群と比較した. BSP 0.25mg/100g/minの持続注入は, 注入開始後20分でBSP排泄率を0.12〜0.13mg/min/100gとし, 以後ほぼ一律であり, その後徐々に低下した. この間BSPの血漿濃度は, 一途上昇を続けた. このようなBSP Tm状態で99mTc HIDAを静注すると, 99mTc HIDAの血漿クリアランスは, 対照に比しおくれるがその差はわずかであった. これに反し, 胆汁内排泄率は, BSP投与ラットで著明に遅延かつ低下した. 60分後の体内臓器分布を検討すると, BSP注入ラットでは, 肝, 腎, 血漿においていずれも対照値より高かった. BSPは99mTcHIDAの肝摂取, 胆汁内排泄 (特に後者) を阻害し, その阻害には, 競合阻害の可能性が示唆される. |

Abstract | [Summary] Plasma clearance and biliary excretion of an iv injected 99mTc-HIDA were compared in control rats and in rats excreting BSP into the bile at a maximal rate (Tm). The iv infusion of BSP (0.25mg/100g/min) for 20 min produced a Tm (0.12 - 0.13mg/100g/min) state which continued for the successive 60 min with a gradual decline in BSP excretion rate. Under this Tm state, plasma clearance of 99mTc-HIDA was slightly but significantly delayed compared with control rats. The biliary recovery of 99mTc-HIDA was considerably decreased in BSP infused rats (60 min recovery, control rats 77.0 +- 5.6% of the dose, mean +- SD, n = 6, BSP rats 33.6 +- 10.0 n = 8). The distribution of 99mTc-HIDA activity at 60 min post injection was higher in the liver, kidneys and plasma in BSP rats. It was suggested that despite the different chemical structure, the presence of BSP in the plasma and liver, inhibits the hepatic uptake and more strongly the biliary excretion of 99mTc-HIDA in rats. |
