Abstract | 「緒言」甲状腺刺激ホルモン (Thyroid stimulating hormone, TSH) は脳下垂体前葉から分泌され, 甲状腺機能を調節する重要なホルモンである. 1959年, Yalow & Bersonによって, 感度, 特異性の優れたradioimmunoassayが開発されたのに引続き, 1967年Odellらは, Condliffeらによって精製されたヒトTSH (HTSH) を用いて, HTSHのradioimmunoassayに成功した. 当初はその供給源が限られていたため一般に普及されなかったが, 最近, 本邦においては測定kitも市販され臨床検査として広く用いられるようになった. 測定用kitそのものの検討については, 我々は既に一部を発表し, 最近諸家によっても報告されつつある. しかしこのようなkitを用いるにあたっては, 従来外国で用いられて来た標識用HTSH, 抗体, 標準HTSHと比較する必要があり, また臨床検査として応用するには詳細な基礎的検討を行わなければならない. そこで我々は以下のような実験を行った. |

Abstract | [Summary] The immunological potency of a reference standard preparation of HTSH 68/38 in terms of HTSH Research Standard A was slightly different according to the labelled HTSH and antisera obtained from different sources, and was estimated as 147×1.31=193mU/ampoule using a HTSH radioimmunoassay Kit "Daiichi" . Reproducibility of the double antibody radioimmunoassay for HTSH using the kit was as follows. The coefficient of variation was 2.2 - 6.5% (within-assay using same lot) , 11.0 - 13.2% (between, assay using same lot) and 6.3 - 10.8% (within-assay using different lot) . No cross reaction was observed with ACTH, GH, LH and FSH. Recovery of HTSH added to serum was 94.8%. Dilution of serum resulted in parallel curves to that obtained for the standard HTSH, when both serum and standard HTSH were diluted with TSH free serum. The minimum detectable concentration and 50% intercept of the standard HTSH were made smaller using two step method. The method was capable of measuring serum HTSH as low as 1〜2μU/ml. |
