Abstract | 「はじめに」α-fetoprotein(以下AFP)は胎児特有の蛋白であり胎児の肝臓において合成され血清中の濃度はGitlin等によれば胎生期の13週に最高濃度となり, その後次第に減少し, 34〜37週で最高値の約20%に減少し, 生後1週月は半減期5日, 2週目以降では半減期3日で減少する. 1963年Abelevはマウスの実験肝癌の血清よりAFPを証明し, 1964年, Tatarinov等が人の肝癌患者の血清からAFPを見出してから肝癌とAFPの関連が多くの研究者により報告されるようになった. 人のAFPは平井等によると分子量64,600, 多糖類含量3%のアルブミンに近い蛋白であり電気泳動法ではアルブミンと, γ-グロブリンの間に易動度を示す. アミノ酸分析の結果は胎児AFPとhepatomaの間に差異を認めないという. AFPの検索は特異的抗血清による抗原抗体反応により行なわれるが, その主な方法としては, (1)Micro Ouchterlony法, (2)Single radial immuno-diffusion法, (3)Immunoelectrosyneresis法(IES法)(4)螢光電気免疫抗散法(FIES法)(5)螢光抗体法, (6)として最高の感度を持つRadioimunoassay法(以下RIA法)があり, 本法は1969年purves, 1971年石井等, 西等により発表され, 1972年にはSeppala等の報告がある. |

Abstract | Alpha fetoprotein has been measured by a radioimmunoassay method. The general procedure is as follows. 0.5ml of buffer solution or 0.4 ml of buffer solution and 0.1ml of hoarse serum is added to a tube, the former is for an unknown sample, the latter for construction of standard curve. Then a known amount of 0.1ml of AFP or a sample containing an unknown amount of AFP is added. In next step, 0.1ml of 125I-AFP and the first antibody are added. The mixture is stirred well and incubated at 4℃ for 24 hours. After an incubation period, 0.1ml of the second antibody is added to the tube and incubated again at 4℃ for 24 hours. Incubation is stopped and the first count is measured. The tube is centrifugated for thirty minutes at 3,000 r. p. m. . The supernant is decanted. The second count is measured. For construction of a standard curve, Ist count/2nd count is plotted for a known concentration of standard alpha fetoprotein. The concentration of standard alpha fetoprotein is 320, 160, 80, 40, 20, 10, 0mμg/ml. The concentration of alpha fetoprotein in an unknown sample is obtained from the observed binding of labelled alpha fetoprotein in the sample by reference to the standard curve. It is possible to detect a level of 1-2mμg/ml of alpha fetoprotein. Alpha fetoprotein in human serum has been considered to be characteristic of the fetal and early neonatal periods and of certain types of malignant tumors. A radioimmunoassay for the determination of alpha fetoprotein is more sensitive than other methods and demonstrates alpha-fetoprotein in serum of patients with hepatitis and liver cirrhosis. In six cases of hepatoma, AFP was demonstrated in all. This commercially available radioimmunoassay kit for determination of alpha fetoprotein(provided by "Dianabot Radioisotope Lab. ") is excellent because of its high sensitivity and presise quantification. |
