Abstract | 「諸言」血中甲状腺ホルモンのうちthyroxine(T4)の測定法として古くから用いられてきた血漿蛋白結合ヨード(PBI)は, 測定法が煩雑で特異性にも問題が多かった. 1964年Murphyによって開発されたcompetitive protein binding analysisは, 一定量の131I(または125I)標識T4と, これに特異的に結合するthyroxine binding protein(TBP)とを反応させ, この反応系に測定すべき非標識T4を競合させた後, 結合-未結合物を分離し, その放射活性を測定するもので, T4とTBPの特異的な結合を利用したすぐれたT4測定法である. その後T4の抽出方法, 結合-未結合物の分離方法などが種々改良され, 多くの変法が報告されている. そのうちで, 近年Mallinckrodt社によって開発されたRes-O-Mat T4法は, アルコールによってT4を抽出し, 結合-未結合物の分離には特殊なresin stripを用いる方法である. |

Abstract | The resin strip technique(Res-O-Mat T4)as a competitive protein binding analysis for determination of serum thyroxine was evaluated. The extraction efficiency factor reached plateau within 10sec. by ethanol extraction and ranged between 75-80%in normal sera. There was no difference between the extraction efficiency factors in hyperthyroid and hypothyroid sera. The standard deviation of duplicate determinations was 0.5μg/dl(T4 iodine)and the coefficient of variations between assays was 11%. The range of concentration of serum thyroxine iodine(mean+-2 S. D. )was 3.0-7.2μg/dl in euthyroid subjects, 5.8-16.0μg/dl in hyperthyroid patients, 0-3.6μg/dl in hypothyroid patients. Oral administration of 6tablets of Telepaque resulted in a significant elevation of PBI but had no effect on T4levels which were determined by Res-O-Mat T4 methcd. |
