Abstract「緒言」1955年Hamolskyは, 131I標識triiodthyronine(131I-T3)赤血球摂取率法をin vitroで行ないうる甲状腺機能検査法として報告した. その後, 1958年, Mitchellは, 赤血球の代わりに, 陰イオン交換樹脂Amberlite IRA-400を用いる131I-T3 resin uptake法を発表した. 更に1960年, Mitchellらは, アメリカAbbott Laboratriesで, 開発されたTriosorb resin spongeを用いることにより好結果を得た. この方法は, 131I-T3 resin sponge uptake testと呼ばれるもので, 現在もっとも広く普及している. Triosorb resin spongeは, polyurethaneに, 陰イオン交換樹脂IRA-400を含有させたもので, T3, T4のみならず, 無機ヨウ素イオンも吸着する. 1965年, 稲垣は, PVF spongeがTriosorb resin spongeに代用しうることを紹介し, 引続き, 1968年, 稲垣, 前田は, 131I-T3摂取率測定にあたり, 測定条件をpH 6.0以下に調整することにより, 一定した値を得ることを発表した.

Abstract[Summary] PVF (Acetalized polyvinyl alcohol) sponge has already been introduced as more useful than the Triosorb resin sponge for determination of T3 uptake rate, and moreover, it has the advantage of strongly adsorbing T3 and T4 with no adsorption of inorganic iodine. This report refers to the mechanism of why the PVF sponge adsorbs T3 and T4. The results were as follows: 1) The binding of T3 to the PVF sponge is believed to be hydrophobic, because non polar solvents eluate T3 far better than polar solvents. 2) High concentrations of urea or guanidine solutions have no effect upon T3 release. Therefore, it is assumed that a hyrogen bond is not involved in the binding. 3) The fact that pH and ionic strength had no influence shows the binding to be non ionic. 4) We investigated the potency octyl alcohol which eluated T3 from the T3 bound PVF sponge under various conditions of temperature and elution time, annd found that about 10% of the bound T3 was rapidly eluated from the sponge within the first minute. The second phase of elution was found to be slow. The latter phase show ed a linear curve by the Arrhenius' plot, which proved the binding mechanism to be thermodynamically uniform. 5) The elution rates of T3 from the T3 bound sponge were determined with alcohol having 1〜9 carbons. Judging from the releasing rate per mol of solvent, butyl alcohol believed to be the best eluating reagent. The facts above mentioned showed that hydrophbic bonds contribute to binding of thyroid hormones to the PVF sponge, which suggests the usefulness of the PVF sponge for separation and determination of non polar substances found in biological materials.
